Research Progress in L-Phenylacetylcarbinol Production by Biosynthesis

Abstract:The research progress in biotransformation of L-phenylacetylcarbinol (L-PAC), including mechanism of biocatalysis, strain screening, biotransformation system, process parameters and downstream processing, was reviewed. Strains belonging to the genera of Saccharomyces and Candida I with high enzyme activity of pyruvate decarboxylase are widely used in biotransformation of L-PAC from benzaldehyde and pyruvate. The formation of by-products and toxicity of substrate significantly influence the production of L-PAC. By strain screening and breeding, suitable strains with high tolerance to benzaldehyde and effective reduction of byproducts can be obtained. By applying the process optimization methodologies of fed-batch culture, process control and process parameter optimization, utilization of cell or enzyme immobilization and adoption of aqueous and organic two-phase system, higher L-PAC production can be achieved.
